Abstract

PT X is a company engaged in the pharmaceutical industry. Every product owned by a pharmaceutical company must have a distribution authorization number (NIE). One of the requirements to obtain an NIE must have a validated and valid analysis model, but the method of analysis owned by PT X has passed its validity period. When it is done revalidation result of resulted accuracy does not fulfill requirement / not valid. Therefore, this research is conducted to find the factors causing the current analysis method is invalid and determine the most suitable method of analysis for testing the dissolution rate of mefenamic acid caplet preparation by anova method. After testing A method is invalid with result of 94,48%. Analysis of factors causing accuracy is not valid by using FMEA is the use of analysis method that is not up to date and the method design is not appropriate, then from that test conducted by conducting a comprehensive study literature so that the proposed B method, C method and get the results 98.15% and 99.31%. Where by comparing the three methods with the anova method obtained the result that the analysis method gives a significant influence on the value of the accuracy of mefenamic acid dissolution rate. Methods B and C can be selected as appropriate methods to replace method A Key Words: Disolution, fishbone diagram, Failure Mode and Effect Analysis (FMEA), ANOVA PT X merupakan perusahaan yang bergerak dibidang industri farmasi. Setiap produk yang dimiliki oleh perusahaan industri farmasi harus mempunyai nomer izin edar (NIE). Salah satu persyaratan memperoleh NIE harus mempunyai Metode analisa yang sudah tervalidasi dan masih berlaku, namun Metode analisa yang dimiliki oleh PT X hampir habis masa berlakunya. Ketika dilakukan revalidasi hasil akurasi yang dihasilkan tidak memenuhi persyaratan/tidak valid. Untuk itu, dilakukan penelitian ini guna mencari faktor – faktor penyebab metode analisa saat ini tidak valid dan menentukan metode analisa yang paling sesuai untuk pengujian kadar disolusi asam mefenamat sediaan kaplet dengan metode anova. Setelah dilakukan pengujian Metode A tidak valid dengan hasil kadar 94,48%. Dilakukan analisa faktor penyebab hasil akurasi tidak valid dengan menggunakan FMEA adalah penggunaan Metode anallisa yang tidak up to date dan desain Metode yang tidak tepat, maka dari itu dilakukan pengujian dengan melakukan study literature menyeluruh sehingga diperoleh usulan Metode B, Metode C dan di dapatkan hasil kadar 98,15% dan 99,31%. Dimana dengan membandingkan ketiga Metode analisa tersebut dengan metode anova didapatkan hasil bahwa Metode analisa memberikan pengaruh yang signifikan terhadap nilai akurasi kadar disolusi asam mefenamic. Metode B dan C dapat dipilih sebagai metode yang sesuai untuk menggantikan metode A Kata Kunci : Disolusi, fishbone diagram, Failure Mode and Effect Analysis (FMEA), ANOVA
